產(chan) 品介紹：The protein encoded by this gene is a lysosomal cysteine proteinase that plays a major role in intracellular protein catabolism. Its substrates include collagen and elastin, as well as alpha-1 protease inhibitor, a major controlling element of neutrophil elastase activity. The encoded protein has been implicated in several pathologic processes, including myofibril necrosis in myopathies and in myocardial ischemia, and in the renal tubular response to proteinuria. This protein, which is a member of the peptidase C1 family, is a dimer composed of disulfide-linked heavy and light chains, both produced from a single protein precursor. At least two transcript variants encoding the same protein have been found for this gene.

免疫熒光技術的實驗步驟：
一、準備好試劑與(yu) 儀(yi) 器：
磷酸鹽緩衝(chong) 鹽水、熒光標記的抗體(ti) 溶液、緩衝(chong) 甘油、搪瓷桶三隻、有蓋搪瓷盒一隻、熒光顯微鏡、玻片架、濾紙、37℃溫箱等。
二、實驗步驟
1.滴加0.01mol/L，pH7.4的PBS於(yu) 待檢標本片上，十分鍾後棄去，使標本保持一定濕度。
2.滴加適當稀釋的熒光標記的抗體(ti) 溶液，使其覆蓋標本，置於(yu) 有蓋搪瓷盒內(nei) ，保溫一定時間以三十分鍾為(wei) 參考。
3.取出玻片，置玻片架上，先用0.01mol/L，pH7.4的PBS衝(chong) 洗後，再按順序過0.01mol/L，pH7.4的PBS三缸浸泡，每缸三到五分鍾，並不停地搖晃振蕩。
4.取出玻片，用濾紙吸去多餘(yu) 水分，但不使標本幹燥，加一滴緩衝(chong) 甘油，以蓋玻片覆蓋。
5.立即用熒光顯微鏡觀察。觀察標本的特異性熒光強度。
